ObjectiveTo summarize recent therapeutic developments for chronic pruritus with a focus on allergic and type 2 inflammatory pathways.Data SourcesLiterature search of PubMed, industry websites, review the ClinicalTrials.gov database.Study SelectionsPeer-reviewed publications public disclosures by relating to pathophysiology therapeutics.ResultsHistamine immunoglobulin E remain primary targets treatment itch in setting urticaria. More recently, blockade immune cell–associated cytokines, including interleukin (IL) 4, IL-13, IL-31, epithelial cell–derived specifically IL-33 thymic stromal lymphopoietin, has is revolutionizing pruritic dermatoses, such as atopic dermatitis prurigo nodularis. Other novel include histamine receptor Janus kinases, ?-opioid receptor, neurokinin 1 phosphodiesterase 4.ConclusionAdvances our understanding neuroimmunology have led identification new rapid development cutting-edge clinical trials. Although incredible advances already been made, continues be an area great unmet need.
